In vitro study | In 3T3 fibroblasts, AR-A014418 inhibited phosphorylation of tau protein at the specific site of GSK3 (serine 396) with an IC50 of 2.7 μm, in turn, N2A cells are protected from death caused by inhibition of the PI3K/PKB signaling pathway. In hippocampal slices, AR-A014418 inhibited neurodegeneration induced by β-amyloid peptide. In NGP cells and SH-5Y-SY cells, AR-A014418 inhibited neuroendocrine markers and inhibited the growth of neuroma cells. In vivo study | In a mouse model of SOD1 protein G93A mutant amyotrophic lateral sclerosis, AR-A014418 (0-4 mg/kg, I. p.) can prolong the occurrence of symptoms, increase muscle vitality, slow down the occurrence of the disease. In addition, AR-A014418 exerts an inhibitory effect on acetic acid and formaldehyde-induced nociception in mice by modulating NMDA and metabotropic receptor signaling pathways and release of TNF-α and IL-1β in the spinal cord.
